SARS-CoV-2 Antigens

In order to mitigate the rising need for in vitro diagnostics in the global COVID-19 scenario, InVivo now offers diverse SARS-CoV-2 proteins in large-scale quantities, which are suitable for the development of highly sensitive IVD kits. Using InVivo’s proprietary manufacturing platforms, different recombinant versions of the viral Spike protein have been transiently expressed in high yielding mammalian HEK cells. These include the two functional units, S1 and S2, and the Receptor Binding Domain (RBD) of the S1 subunit that directly engages with host cells via the human receptor Angiotensin Converting Enzyme 2 (ACE2).

To cater to larger demand of long-term bulk quantities in the gram scale, InVivo has also initiated development of several stable CHO cell lines.

Monoclonal antibody production

Our in vitro production in super spinner systems or bioreactors (2 L to 100 L) using high-cell density fermentation, occurs exclusively under serum-free conditions. For that purpose we use our proprietary medium ISF-1 to allow cost-effective production processes and high product yields.

Antibodies regularly ordered in larger quantities (> 1g), qualify for our special antibody-on-demand service. This offer reduces your costs and allows for pre-scheduled deliveries that suit your needs. You just define the timeline of your demand, say 24 months, choose a batch size starting from 100 mg per call-off and select your preferred dates of delivery. We take care of the rest.

Mouse hybridoma development

With passion and know-how, InVivo has successfully realized more than 500 hybridoma development projects to date. The entire project, including mice immunization, cell fusion, screening, cloning and initial characterization of the developed monoclonal antibodies, usually takes around 22 weeks.

For immunization ~ 5 mg of antigen (protein, peptide or hapten) is needed. In case the antigen (protein) is not available yet, InVivo can produce the protein of interest via expression in E. coli or via transient gene expression in HEK cells.

Recombinant antibody production

At InVivo, your recombinant antibody production is in reliable hands. Recombinant antibodies for target screening, antigen production or drug discovery can be produced either by transient expression or from your own stable cell line in small to large scales. Transient expression is performed using our in house system InVEST. InVEST guarantees an approximately 80 fold increase in yield compared to commercially available systems, at reduced costs. Within the last 20 years, our experts have acquired significant expertise by producing all kinds of antibodies from humans, rat or mouse.

Stable CHO cell line development

Through intensive research and process development, InVivo optimized the well-established CHO-IS cell line and adapted it to a high-performance chemically-defined, animal component-free medium.

Moreover, novel expression vectors were designed which are compatible to the PiggyBac system, a well-established tool for transposon-mediated active gene transfer. Using these new vectors, high protein yields and a substantially reduced timeline from transfection to stable CHO cell pools could be achieved.

The combined action of highly optimized processes for the efficient selection of best producing cell pools or high-producing monoclonals, helps achieve high-cell-density fermentation processes in standard bioreactors. This enables cost-optimal production of large quantities of target proteins.

Recombinant proteins from mammalian expression system

Transient Gene Expression (TGE) is a commonly used method for the production of recombinant antibodies and proteins in mammalian cells within a short time.

However, optimal results can only be achieved, if all components of the TGE system are carefully matched and optimized.

Within a few years of research and development, InVivo established an optimized system for the production of mammalian-derived proteins via transient gene expression in HEK cells. This system is called InVEST and it combines an optimized host cell line with sophisticated vector design as well as effective large-scale plasmid preparation and customized cell culture media.

Recombinant proteins from E.coli expression system

Using E. coli as expression system for the production of recombinant proteins has several advantages: a shorter timeline for the entire process from cloning to protein recovery, inexpensive production processes, high protein yields and highest flexibility in production scale.

Smaller peptides and proteins without posttranslational modifications, like antigens, bacterial proteins, hormones, pro-hormones, tumor markers or cytokines, can be expressed in bacterial cells.

Every E. coli protein production project includes cDNA design and efficient codon optimization for expression in bacterial cells.
